I think it's in She Wore a Yellow Ribbon, but I wouldn't swear to it.

But on second thoughts, it could be 2nd lieutenant O'Rourke when sent out with a detail of four men to look for a wagon in Rio Grande. Both films are part of the "cavalry trilogy" of John Wayne.
